ABSTRACT We conducted experimental feeding using 3 feeding methods (pile, spread, trough) and 2 quantities (rationed, ad libitum) of shelled corn to compare deer activity and behavior with control sites and evaluate potential direct and indirect transmission of infectious disease in white-tailed deer (Odocoileus virginianus) in central Wisconsin, USA. Deer use was higher at 2 of the feeding sites than at natural feeding areas (P ≤ 0.02). Deer spent a higher proportion of time (P < 0.01) feeding at pile (49%) and spread (61%) treatments than at natural feeding areas (36%). We found higher deer use for rationed than ad libitum feeding quantities and feeding intensity was greatest at rationed piles and lowest at ad libitum spreads. We also observed closer pairwise distances (≤0.3 m) among deer when corn was provided in a trough relative to spread (P=0.03). Supplemental feeding poses risks for both direct and indirect disease transmission due to higher deer concentration and more intensive use relative to control areas. Concentrated feeding and contact among deer at feeding sites can also increase risk for disease transmission. Our results indicated that restrictions on feeding quantity would not mitigate the potential for disease transmission. None of the feeding strategies we evaluated substantially reduced the potential risk for disease transmission and banning supplemental feeding to reduce transmission is warranted.  相似文献

We have analysed the proteinase profiles of 11 species from 7 different genera of trypanosomatids by in situ detection of enzyme activities on SDS-PAGE gels containing co-polymerized gelatin as substrate, and the use of specific proteinase inhibitors. Our survey indicates that while cysteine- and metalloproteinases are distributed ubiquitously among trypanosomatids, there are marked differences between the enzyme profiles from the monogenetic (Crithidia, Herpetomonas, Leptomonas) and digenetic (Trypanosoma, Endotrypanum, Phytomonas, Leishmania) species. The detected metalloproteinase activities, ranging in size from 50–100 kDa, partitioned into the detergent-phase after Triton X-114 extraction, while most of cysteine proteinases, of three distinct molecular mass ranges (30–50 kDa, 80–100 kDa and 116–205 kDa), partitioned into the aqueous phase. Thus, within this group of organisms, the metalloproteinase activities seem to be predominantly membrane-associated proteins. We also show that the plant parasites of the genus Phytomonas exhibit a distinctive cysteine proteinase profile that might be exploited further as a criterion for taxonomy of the genus.  相似文献

1. The number of hosts attacked as a function of host density is considered to be an important characteristic of parasitoid behaviour and is used to estimate key parameters such as handling time and ‘instantaneous rate of discovery’. However, little has been done to validate functional response models by direct observation of parasitoid oviposition behaviour. 2. Tamarixia radiata is the most promising parasitoid for biological control attacking Diaphorina citri. Mass rearing and augmentative release seen as a potential strategy for suppression of D. citri has been documented in abandoned citrus, residential areas, and organic groves. Nevertheless, parasitism rates in culture and in the field are only moderate, leading to questions about oviposition behaviour in response to host density. 3. Behaviours of gravid T. radiata females presented with susceptible host instars were categorised and documented by direct observation for 30 min and by camera recordings made over 12 h. Frequency of searching and antennating increased with host density during the 30 min. Probing rejection rates and search duration increased significantly with host density over 12 h. These factors resulted in significantly lower fecundity than expected, possibly due to host mark‐mediated deterrence within the small searching area. Females took approximately 3.6–4.2 min to probe and parasitise a host regardless of host density and exposure duration. These results were markedly different from the 52.2 min estimated from the functional response equation. 4. Further experiments are required to assess the range and persistence of the putative host‐marking pheromone, and to better understand the relationship between functional response parameters and actual behaviour.  相似文献

Both island-biogeographic (dynamic) and niche-based (static) metapopulation models make predictions about the distribution and abundance of species assemblages. We tested the utility of these models concerning such predictions for terrestrial vascular plants using data from 74 landscapes across the globe. We examined correlations between species frequency and local abundance and shapes of the species frequency distribution. No data set met all of the predictions of any single island-biogeographic metapopulation model. In contrast, all data sets met the predictions of the niche-based model. We conclude that in predicting the distribution of species assemblages of plants over scales greater than 10–1 km, niche-based models are robust while current metapopulation models are insufficient. We discuss limitations in the assumptions of the various models and the types of empirical observations that they will each have to deal with in further developments.  相似文献

THYMUS-derived, T lymphocytes and bursa-equivalent B lymphocytes cooperate in the initiation of the humoral antibody response of mammals to a variety of antigens1–5. The B cells are antibody-forming-cell precursors (AFCP) and the T cells are helper cells which serve to augment the antibody response produced by the precursor cells6. Mitchison and his colleagues4,5,7 have shown that the interaction between carrier and hapten-primed cells in the adoptive secondary antibody response to hapten-protein conjugates is an example of cooperation between T and B lymphocytes respectively.  相似文献

SYNOPSIS. When the synthetic polyribonucleotides polyinosinic acid-polycytidylic acid (poly I poly C) and polyadenylic acid-polyuridylic acid (poly A poly U) were tested against mice infected with varying numbers of Trypanosoma congolense the results varied with the method of passage of trypanosomes in mice. Thus, when 100 flagellates were passaged every 7th day and experiments were initiated with these trypanosomes from mice on the 7th day of their infection, the protective effects of poly I poly C and poly A poly U apparently varied independently of each other as assayed by the mean parasitemias and cumulative mortalities of infected mice. Poly I poly C-resistant and poly I poly C-susceptible variants (“R” and “S”, respectively) were isolated and maintained in mice by passage of 106 trypanosomes every 4th day. Mice infected with these variants responded consistently to poly I poly C and poly A poly U injections in that mice infected with the “R” variant showed no response to either polyribonucleotide but those infected with the “S” variant consistently had a decrease in mean parasitemias and cumulative mortality when treated with poly I poly C, but not with poly A poly U. Using mice infected with the “S” variant, the protective effect of poly I poly C was dose-dependent and best protection was afforded when 1st injections of poly I poly C were given around the time of infection of the mice. The protective effects of the synthetic polyribonucleotides used in these experiments are probably due to their immunologic enhancing capacities and not to interferon. To support this, injections of Newcastle disease virus, a strong inducer of interferon in mice, did not protect against T. congolense in mice. It was not possible to determine whether serum from poly I poly C-treated mice had a greater neutralizing effect upon trypanosomes in vitro than serum from saline-treated mice since any effect of antibody was masked by a naturally occurring inhibitor in normal mouse serum which was reduced during infection. The protective effects of poly I poly C in T. congolense-infected mice were reversed by treatment with cyclophosphamide. This strong immunosuppressant, however, could not reverse the protective effects of poly I poly C against mice infected with Semliki Forest virus, strongly suggesting that the protective mechanisms stimulated by poly I poly C in these 2 infections were different. The response of mice infected with the “R” and “S” variants of T. congolense to synthetic polyribonucleotides is discussed in relation to antigenic variation of trypanosomes.  相似文献

Male fiddler crabs (Genus Uca) employ both visual and acousticalsignals to attract females for mating. In U. pugilator and severalother American species, the males attract females during theday first by waving, then by producing sounds just within theirburrows. At night, the males produce sounds at low rates, butwhen touched by a female, they increase their rate of soundproduction. In the European species, U. tangeri, many elements of courtshipare similar to those in U. pugilator, but two types of soundsare produced. One of these, the short drumwhirl, appears tosubstitute for waving when the male is temporarily obscuredfrom the female during his diurnal courtship activities. Thelong drumwhirl is used under different circumstances. The acoustical responses of a male to a female influence thecourtship behavior of other males in the area. When sounds fromstimulated males are played back to test males during the day,their lates of waving increase. At night, the playbacks elicitincreases in rates of sound production. The influence of tidal oscillations, temperature, and lightcycles on the behavior of males is discussed. Courtship activities of aquatic crabs are compared to thoseof terrestrial Brachyura. In aquatic forms, courtship may beabsent or, if present, does not involve elaborate signallingby the male. Chemical or visual cues at close range are themost important stimuli. In several genera of terrestrial crabs,visual signalling for prolonged periods is common, and soundsare often emitted by males to "call" females from their burrowsto the surface for mating. Some of the factors that may accountfor differences in courtship activities in aquatic and terrestrialspecies are discussed.  相似文献

The arrangement of experiments for intercomparison of a number of virus samples is discussed and the use of randomised blocks or Latin squares, combined in certain cases with half-leaf comparisons between all samples or against a standard, is recommended.
Improvements in the ground-glass spatula method of inoculation have been suggested.
Provided there is sufficient inoculum to cover the leaf when the spatula is rubbed over it, the amount, of inoculum makes no difference to the number of lesions produced.
The conditions to which test plants are subjected shortly before inoculation were found to influence the number of lesions produced by a given inoculum.
The number of lesions produced by otherwise similar inocula is influenced by their pH value and electrolyte content. The optimum pH . range for the virus of tomato spotted wilt is very limited (roughly pH 6.0–8.5). Tobacco mosaic virus in a potassium phosphate buffer at a pH value about 7 produced the maximum number of lesions in the concentration range 0.05-0.2 M . It is recommended that for most quantitative work viruses should be buffered at a definite pH value.
When effects due to varying pH value and electrolyte content were excluded, the virus of tomato spotted wilt was still found to be inactivated by certain oxidising agents and preserved by certain reducing agents.  相似文献

Strips of epidermal tissue bearing hair cells were removed fromtomato stems. The lower surface exposed by the stripping wasinoculated with tobacco mosaic virus and the progress of infectionof the hair cells was followed microscopically and by extractionof infectivity. Shortly before infectivity could be extractedfrom the hair cells, the nuclei were observed to have migratedtowards the tran sverse wall of the basal hair cell nearestthe epidermis, apparently the site at which infection of thecell commences. After infection was apparent, the staining characteristicsof the nuclei, before and after RNase and DNase treatment, suggestedan accumulation of RNA. A granular body, different from X-bodiesmaintained a close association with the nucleus of TMV infectedhair cells, and was found to be rich in RNA, suggesting thatviral RNA of the nucleus may have been entering the cytoplasmvia the granular body.
